Photoactivation: the light-driven assembly of the water oxidation complex of photosystem II

Resumo

A oxidação fotossintética da água é catalisada pelo aglomerado Mn₄CaO₅ do fotossistema II. A montagem do aglomerado Mn₄O₅Ca requer luz e envolve um processo sequencial denominado fotoativação. Esse processo utiliza a separação de cargas do centro de reação fotoquímica e o ambiente de coordenação fornecido pelas cadeias laterais de aminoácidos da proteína para oxidar e organizar os íons de manganês que chegam, formando o aglomerado metálico ligado por oxo, capaz de oxidar a H₂O. Embora a maioria dos aspectos desse processo de montagem permaneça pouco compreendida, avanços recentes na elucidação da estrutura cristalina do complexo PSII de cianobactérias totalmente montado ajudam na interpretação da rica história de experimentos projetados para entender esse processo. Além disso, novos conhecimentos sobre a estrutura e estabilidade dos íons constituintes do aglomerado Mn₄CaO₅ podem orientar experimentos futuros. Aqui, consideramos a literatura e sugerimos possíveis modelos de montagem, incluindo um que envolve um único local de oxidação de Mn²⁺ para todos os Mn, mas que exige realocação de íons.



Abstract

Photosynthetic water oxidation is catalyzed by the Mn4CaO5 cluster of photosystem II. The assembly of the Mn4O5Ca requires light and involves a sequential process called photoactivation. This process harnesses the charge-separation of the photochemical reaction center and the coordination environment provided by the amino acid side chains of the protein to oxidize and organize the incoming manganese ions to form the oxo-bridged metal cluster capable of H2O-oxidation. Although most aspects of this assembly process remain poorly understood, recent advances in the elucidation of the crystal structure of the fully assembled cyanobacterial PSII complex help in the interpretation of the rich history of experiments designed to understand this process. Moreover, recent insights on the structure and stability of the constituent ions of the Mn4CaO5 cluster may guide future experiments. Here we consider the literature and suggest possible models of assembly including one involving single Mn2+ oxidation site for all Mn but requiring ion relocation.
