The Role of Abscisic Acid Signaling in Maintaining the Metabolic Balance Required for Arabidopsis Growth under Nonstress Conditions

O ácido abscísico (ABA) é um hormônio vegetal que regula uma gama diversificada de processos celulares e moleculares durante o desenvolvimento e em resposta ao estresse osmótico. Em Arabidopsis thaliana, três proteínas quinases 2 relacionadas a Sacarose não fermentável 1 (SnRK2s), SRK2D, SRK2E e SRK2I, são reguladores positivos chave envolvidos na sinalização de ABA cujos substratos foram bem estudados. Além da tolerância reduzida ao estresse hídrico, o mutante srk2d srk2e srk2i mostra fenótipos de crescimento anormais, como um número aumentado de folhas, sob condições não estressantes. No entanto, permanece incerto se, e se sim como, a sinalização de ABA mediada por SnRK2 regula o crescimento e o desenvolvimento. Aqui, mostramos que o perfil de metabólitos primários de srk2d srk2e srk2i cultivado sob condições não estressantes foi consideravelmente diferente do de plantas selvagens. As mudanças metabólicas observadas em srk2d srk2e srk2i foram semelhantes às de um mutante de biossíntese de ABA, aba2-1, e ambos os mutantes mostraram uma taxa de emergência de folhas maior que o tipo selvagem. Consistente com as quantidades aumentadas de citrato, experimentos de marcação isotópica revelaram que a respiração através do ciclo do ácido tricarboxílico foi intensificada em srk2d srk2e srk2i. Esses resultados, juntamente com dados de transcriptoma, indicam que as SnRK2s envolvidas na sinalização de ABA modulam o metabolismo e o crescimento foliar sob condições não estressantes ao ajustar finamente o fluxo através do ciclo do ácido tricarboxílico.
Abstract
Abscisic acid (ABA) is a plant hormone that regulates a diverse range of cellular and molecular processes during development and in response to osmotic stress. In Arabidopsis (Arabidopsis thaliana), three Suc nonfermenting-1-related protein kinase2s (SnRK2s), SRK2D, SRK2E, and SRK2I, are key positive regulators involved in ABA signaling whose substrates have been well studied. Besides reduced drought-stress tolerance, the srk2d srk2e srk2i mutant shows abnormal growth phenotypes, such as an increased number of leaves, under nonstress conditions. However, it remains unclear whether, and if so how, SnRK2-mediated ABA signaling regulates growth and development. Here, we show that the primary metabolite profile of srk2d srk2e srk2i grown under nonstress conditions was considerably different from that of wild-type plants. The metabolic changes observed in the srk2d srk2e srk2i were similar to those in an ABA-biosynthesis mutant, aba2-1, and both mutants showed a higher leaf emergence rate than wild type. Consistent with the increased amounts of citrate, isotope-labeling experiments revealed that respiration through the tricarboxylic acid cycle was enhanced in srk2d srk2e srk2i. These results, together with transcriptome data, indicate that the SnRK2s involved in ABA signaling modulate metabolism and leaf growth under nonstress conditions by fine-tuning flux through the tricarboxylic acid cycle.
