The cold-stress responsive gene DREB1A involved in low-temperature tolerance in Xinjiang wild walnut

Resumo

Contexto: Baixas temperaturas têm o potencial de ser um problema sério para as plantas e podem afetar negativamente o crescimento e desenvolvimento normais das nogueiras. DREB1/CBF (Proteína de Ligação ao Elemento Responsivo à Desidratação 1/Fator de Ligação à Repetição C), um dos fatores de transcrição mais diretos em resposta ao estresse de baixa temperatura, pode melhorar a resistência das plantas a baixas temperaturas regulando seus genes funcionais. No entanto, poucos estudos foram conduzidos em nogueira. A nogueira selvagem de Xinjiang é uma planta selvagem rara encontrada na China, com um grande número de genes de características excelentes, e é mais resistente do que as nogueiras cultivadas em Xinjiang.

Métodos: Neste trabalho, identificamos todos os membros DREB1 do genoma da nogueira e analisamos seus níveis de expressão em diferentes tecidos e durante o estresse de baixa temperatura na nogueira selvagem de Xinjiang. O gene JfDREB1A da nogueira selvagem de Xinjiang foi clonado e transformado em Arabidopsis thaliana para verificação funcional.

Resultados: Havia cinco fatores de transcrição DREB1 no genoma da nogueira. Entre eles, o nível de expressão relativa do gene DREB1A foi significativamente maior do que outros membros nos diferentes tecidos (raiz, caule, folha) e foi imediatamente regulado positivamente sob estresse de baixa temperatura. A superexpressão do gene JfDREB1A aumentou as taxas de sobrevivência das linhagens transgênicas de Arabidopsis, principalmente por meio da manutenção da estabilidade da membrana celular, diminuindo a condutividade elétrica e aumentando as atividades de enzimas antioxidantes, incluindo superóxido dismutase (SOD), peroxidase (POD) e catalase (CAT). Além disso, os níveis de expressão de genes indutíveis pelo frio como AtKIN1, AtERD10, AtRD29A, AtCOR15A e AtCOR47 foram significativamente aumentados. Esses resultados mostraram que o gene JfDREB1A pode desempenhar um papel importante na resposta ao estresse pelo frio da nogueira selvagem de Xinjiang. Este estudo contribui para a compreensão do mecanismo molecular da resposta da nogueira selvagem de Xinjiang ao estresse de baixa temperatura e será benéfico para o desenvolvimento de cultivares de nogueira com maior resistência ao frio.



Abstract

Background: Low-temperatures have the potential to be a serious problem for plants and can negatively affect the normal growth and development of walnuts. DREB1/CBF (Dehydration Responsive Element Binding Protein 1/C-repeat Binding Factor), one of the most direct transcription factors in response to low-temperature stress, may improve the resistance of plants to low-temperatures by regulating their functional genes. However, few studies have been conducted in walnut. The Xinjiang wild walnut is a rare wild plant found in China, with a large number of excellent trait genes, and is hardier than cultivated walnuts in Xinjiang.
Methods: In this work, we identified all of the DREB1 members from the walnut genome and analyzed their expression levels in different tissues and during low-temperature stress on the Xinjiang wild walnut. The JfDREB1A gene of the Xinjiang wild walnut was cloned and transformed into Arabidopsis thaliana for functional verification.
Results: There were five DREB1 transcription factors in the walnut genome. Among them, the relative expression level of the DREB1A gene was significantly higher than other members in the different tissues (root, stem, leaf) and was immediately un-regulated under low-temperature stress. The overexpression of the JfDREB1A gene increased the survival rates of transgenic Arabidopsis lines, mainly through maintaining the stability of cell membrane, decreasing the electrical conductivity and increasing the activities of antioxidant enzymes including superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). Additionally, the expression levels of cold-inducible genes like AtKIN1, AtERD10, AtRD29A, AtCOR15A and AtCOR47, were significantly increased. These results showed that the JfDREB1A gene may play an important role in the response to cold stress of the Xinjiang wild walnut. This study contributes to our understanding of the molecular mechanism of the Xinjiang wild walnut’s response to low-temperature stress and will be beneficial for developing walnut cultivars with improved cold resistance.
