Humic acids and Herbaspirillum seropedicae change the extracellular H+ flux and gene expression in maize roots seedlings

Resumo

Contexto: O uso de bioestimulantes de origem vegetal para aumentar a produtividade das culturas e melhorar a proteção tem crescido em popularidade. Anteriormente, produzimos um inoculante combinando ácidos húmicos com bactérias promotoras de crescimento de plantas e observamos um efeito significativo na produtividade das culturas. No entanto, estudos de eletrofisiologia que elucidam os mecanismos responsáveis pela maior absorção de nutrientes e água são escassos e geralmente restritos a modelos in vitro.

Materiais e métodos: Neste estudo, uma suspensão de ácidos húmicos isolados de vermicomposto, juntamente com Herbaspirillum seropedicae, foi aplicada a plântulas de milho. Medimos o fluxo de H⁺ nas raízes com um sistema de sonda vibrante seletiva a íons. Além disso, foi medida a transcrição da ATPase H⁺ da membrana plasmática, de aquaporinas e de transportadores de nitrato de alta ou baixa afinidade.

Resultados: A inoculação ativou o fluxo extracelular de H⁺, alterando o pH e o potencial de membrana das células da raiz do milho e modificando o potencial eletroquímico gerado pela P–H⁺-ATPase em nível bioquímico e molecular. A superexpressão de aquaporinas também foi observada; no entanto, os transportadores de nitrato foram reprimidos pelos inoculantes.

Conclusão: Demonstramos um aumento no efluxo de H⁺ em plântulas de raiz de milho inoculadas com H. seropedicae e mudanças concomitantes na voltagem da membrana. A inoculação de H. seropedicae na presença de ácidos húmicos diminuiu o fluxo de H⁺ e a acidificação da superfície sem alterações no nível de transcrição das aquaporinas. No entanto, o fluxo de H⁺ nas plântulas inoculadas (com ou sem ácidos húmicos) foi maior em relação às plantas controle. Esses resultados corroboram a maior eficiência hídrica e nitrogenada em plantas inoculadas com ácidos húmicos e H. seropedicae.



Abstract

Background
The use of plant-based biostimulants to increase crop yield and enhance protection has grown in popularity. We previously manufactured an inoculant combining humic acids with plant growth-promoting bacteria and observed a significant effect on crop yields. However, electrophysiology studies elucidating the mechanisms responsible for enhanced nutrient and water uptake are scarce and are generally restricted to in vitro models.

Materials and methods
In this study, a suspension of humic acids isolated from vermicompost along with Herbaspirillum seropedicae was applied to maize seedlings. We measured the root H+ flux with an ion-selective vibrating probe system. Furthermore, the transcription of plasma membrane H+ ATPase, aquaporin and high- or low-affinity nitrate transporters was measured.

Results
Inoculation activated the extracellular H+ flux, thus changing the pH and membrane potential of maize root cells and altering the electrochemical potential generated by P–H+-ATPase at the biochemical and molecular level. The overexpression of aquaporins was also observed; however, nitrate transporters were repressed by the inoculants.

Conclusion
We demonstrate an increase in the H+ efflux in maize root seedlings inoculated with H. seropedicae and concomitant changes in membrane voltage. The inoculation of H. seropedicae in the presence of humic acids decreased the H+ flux and surface acidification without changes in the aquaporin transcription level. However, the H+ flux in root seedlings in inoculated plants (with or without humic acids) was larger in respect to control plants. These results support the increased water and nitrogen efficiency in plants inoculated with humic acids and H. seropedicae.
